Semax.

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) studied in models of neurotrophic signaling and gene transcription. An analogue of an ACTH fragment extended by Pro-Gly-Pro, it is described in the preclinical publications below. How the semax peptide works: the BDNF and NGF neurotrophins

Semax derives from fragment 4-7 of ACTH (adrenocorticotropic hormone), to which a Pro-Gly-Pro tail gives increased resistance to peptidases. Unlike native ACTH, it has no significant effect on the hypothalamic-pituitary-adrenal axis: its research interest lies in the central nervous system, where it modulates neurotrophin expression.

Work by Dolotov and colleagues established that Semax rapidly increases expression and phosphorylation along the BDNF/trkB pathway in the rodent hippocampus, and raises BDNF protein levels in the basal forebrain. Further transcriptomic studies showed activation of the genes encoding neurotrophins (including NGF, nerve growth factor) and their receptors after experimental ischaemic injury. This trophic modulation sits at the centre of current research protocols: synaptic plasticity, neuroprotection in cerebral ischaemia models, and working memory in behavioural models.

A genome-wide analysis (Medvedeva et al., 2014) additionally showed that Semax affects the expression of genes tied to the immune and vascular systems in focal cerebral ischaemia — which makes it a tool for studying the interaction between neuroinflammation and post-ischaemic recovery. For laboratories it is therefore a reference peptide for exploring neurotrophic cascades without confounding the results with corticotropic hormonal activity.
