Selank.

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic heptapeptide developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. An analogue of tuftsin extended by a Pro-Gly-Pro sequence, it is studied in models covering GABAergic neurotransmission and cytokine modulation. How Selank works: the GABAergic pathway and IL-6 modulation

Selank inherits its immunomodulatory properties from tuftsin (Thr-Lys-Pro-Arg), to which the Pro-Gly-Pro tail adds greater stability against peptidases. Its research profile is organised around two main axes: GABAergic neurotransmission and the regulation of inflammatory cytokines.

On the GABAergic side, the reference study by Volkova et al. (2016, Frontiers in Pharmacology) showed that Selank alters the expression of dozens of genes involved in GABAergic neurotransmission — GABA-A receptor subunits, transporters, and GABA synthesis enzymes — within an hour of administration in rodents. Rather than direct benzodiazepine-style binding, this appears to be allosteric modulation of the system, which is what makes research models of selank and anxiety interesting: an experimental anxiolytic profile without the sedation or tolerance characteristic of benzodiazepines in those models.

On the immune side, Russian preclinical and clinical work has reported a normalisation of the Th1/Th2 cytokine balance and suppression of IL-6 gene expression in experimental stress and anxiety models. Selank also influences enkephalin metabolism by inhibiting the enzymes that degrade them. This dual signature — GABA plus cytokines such as IL-6 — makes it a strong tool for studying the interface between the nervous and immune systems.
